I like PVE content in MMOs. I like the feeling of working together with a group of friends to overcome a challenge. In particular, the type of challenge presented by boss fights. Most MMOs have very similar combat though, it is real-time, 3rd-person, and based on the holy trinity of tank-dps-healer.

What I'm looking for is a game that recreates the feel of PVE group content. It can be single player or coop, real-time or turn-based. Any genre is fine as long as it isn't derivative of WoW.

Republic Commando would be a good example.

What about all those co-op FPS? Though I don't know any focused on bossfights, even if they include one the challenge is mainly getting through a level with the boss being just the final challenge or a challenge spike

There's Evolve I guess

DotA 2 is/has rolled out custom map making so there's going to be some of those custom map kind of experiences like there was with WC3 and Brood War.
