Actually, there's a mission to conquer Rome once you own South Italy, which is particularly awesome since Byzantium is an Orthodox nation. Unlike Catholic nations, you won't be consistently spammed with events to re-form the Papal State, and an Orthodox nation controlling Rome gets +1 missionaries per year and a yearly prestige bonus. (Same goes for controlling Judea/Jerusalem, which you don't get a mission for, but which is close enough to Byz to be worth considering.)
There's also a mission to destroy (conquer) Venice, which gives you free cores on Venice and Treviso. And the same for Sicily (although that's not technically on the Iberian peninsula). Byzantium gets awesome missions.
